Application Notes - Pharmaceutical

Sometimes it’s tough to envision the applicability of ChemImage technology outside the laboratory or field environment. The following collection of application notes contain real-world discussions about the FALCON II™ Raman Chemical Imaging and the CONDOR Wide-field Hyperspectral Imaging technology. Discrimination of Acetaminophen Polymorphs Using Raman Chemical Imaging
(PDF 437 KB)
Screening for polymorphs is crucial for drug research and development since the crystal form of the active pharmaceutical ingredient (API) directly correlates with its pharmacokinetic properties. Raman spectroscopy and chemical imaging has a vast potential to identify and control polymorphic purity of pharmaceutical compounds.

Identification of Drug Forms in Solid Solutions Using Wide-field Raman Chemical Imaging (PDF 342 KB)
Characterizing the drug form is critical for designing products with an increased rate of drug dissolution, studying drug-excipient compatibility and stability, and detection of polymorph forms. Wide-field RCI may be used to distinguish drug forms in polymer solutions and dispersions providing representative information from a large sampling area with sub-micron spatial resolution and high spectral resolution.

Ingredient-Specific Particle Sizing of Aerosolized Nasal Spray* Using Wide-Field Raman Chemical Imaging (PDF 843 KB)
Creating the correct nasal formulation is both challenging and critical. Nasal sprays formulated as suspensions typically contain micronized active pharmaceutical ingredient (API) in the presence of multiple excipient materials.

Characterization of A Controlled Release Bead Using Wide-Field Raman Chemical Imaging (PDF 387 KB)
A pharmaceutical controlled release bead with an unusual release profile was submitted for analysis. After the bead was microtomed, Raman spectra, brightfield images and Raman chemical images were obtained using a FALCON II™ Wide-Field Raman Chemical Imaging System with 532 nm excitation. ChemImage XpertTM was used to visualize and measure the thickness of the two coating layers.

Measuring Content Uniformity In Pharmaceutical Tablets Using Wide-Field Raman Chemical Imaging and Multivariate Image Processing (PDF 505 KB)
This note discusses analysis of an over-the-counter (OTC) pharmaceutical tablet which exhibits compositional heterogeneity between the active ingredient (aspirin) and the excipient matrix within the sample. The intact tablet appears homogeneous on a macro or microscopic scale when using conventional contrast enhancement since both the active and excipient materials are white powders and cannot be readily differentiated using optical microscopy.

Monitoring Droplet Size Distribution in a Complex Emulsion System Using Wide-Field NIR Chemical Imaging (PDF 440 KB)
A thin layer of an over-the-counter (OTC) cream was prepared on a microscope slide. Brightfield transmission images and NIR chemical images were obtained using a FALCON II™ Wide-Field Raman Chemical Imaging System employing an optional NIR camera. ChemImage XpertTM software was used to process the imaging data and generate droplet size distribution statistics for 476 droplets with a mean size of 3.2 ± 1.9 μm.
